Plasma proteins as a predictor of chronological age in people living with HIV: a cross-sectional study
نویسندگان
چکیده
BackgroundAgeing is associated with an impaired immune response and shares many physiological alterations observed in HIV chronic infection; therefore, we hypothesised that people living exhibit a premature ageing phenotype, reflected by age-related changes the plasma proteome.MethodsWe profiled proteome two cohorts of virally suppressed HIV, 2000HIV cohort (n=65; median age 53 years [IQR 45–60]; 91% male [n=597] 8% female [n=59]) 200HIV (n=218; 52 [45–59]; [n=199] 9% [n=19]), 100 age-matched sex-matched without 200FG (n=100; 50 [38–59]; 75% [n=75] 25% [n=25]). Patients were recruited from following Dutch Treatment Centres: Radboudumc (Nijmegen), ErasmusMC (Rotterdam), Onze Lieve Vrouwe Gasthuis (Amsterdam), Elisabeth-TweeSteden Ziekenhuis (Tilburg). Clinical information was collected all participants using standard questionnaires interviews on lifestyle, health, clinical symptoms, samples blood, urine, saliva stool obtained for further analytical measurements. We characterised proteins are chronological explored extent to which predicts or HIV. Using multiplex proximity extension assay (Olink Proteomics), relative abundance 1472 assessed across four different panels (inflammatory, cardiometabolic, neurology, oncology). The used build prediction model age, based elastic net regression common proteomic signature between HIV.FindingsThe number significantly 569 2000 cohort, 322 200 101 (adjusted p<0·05). 78 some these (LTBP2, EDA2R, WNT9A, NEFL, SCARF2, KLK4, GDF15, CXCL14, HAVCR1, GFAP) involved immune-related processes secretory senescence-associated phenotype. correlation 0·92 200FG, 0·82 0·8 (p<0·001). According this proteomic-age predictive model, showed increased inflammatory compared HIV.InterpretationThis targeted proteomics analysis This could predict warrants investigation its role development comorbidities disease severity.FundingViiV Healthcare.
